Morphologic Canges of Central Nervous System, Liver and Kidney After Intraperitoneal Lead Indection in rat

Abstract
Lead is one of the representative hazardous materials which have the effect on hematopoiectic system, nervous system and gastrointestinal tract by environmental and occupational exposures. For the evaluation of the effects of lead exposure on nervous system. Liver and kidney, the author observed morphologic changes of neurons that have tyrosine hydroxylase (TH) which is the precursor for the synthesis of catecholamine and histopathologic findings of liver and kidney. Lead acetate was intraperitoneally injected daily into the male rats(Sprague-Dawley) with high (0.1mg/g) and low dosages(0.01mg/g), and the results were observed on the first, second, third, fifth. And fourteenth day. The concentration of lead in whole blood was analyzed with atomic absortion spectrophotometry. The number and the staining intensity of TH containing nerve cells sampled from the segment of substantia nigra and locus cerulus were observed by immunohistochemistry. Histopathologic findings of liver and kidney were observed by hematoxylin & neosin staining with light microscope.
The results are as follows:
1. The concentration of lead in whole blood was increased by the increased dose of lead acetate and there was no difference between high and low dosage groups.
2. TH containing cells were intensely stained in the high dosage group compared with the low dosage group, and the number and the staining intensity of TH containing cells were markedly increased in the sample of fifth day. After the fifth day the cell number and the staining intensity were decreased and locus cerulus showed more decreasing tendency.
3. There was moderate degree of necrosis and inflammation on the fourteenth day. Fibrosis and inclusion body in live were not observed. There were no specific findings for the low dosage group.
4. In the histopathologic findings of kidney there were neutrophile and eosinophile infiltration around glomerular capillaries, loss and atrophy of tubular epithelial cell, mild interstitial fibrosis and inflammation, but no vascular changes and inclusion bodies were observed. And there were also no specific findings for the low dosage group.
